Abstract
Asaparagine-linked glycosyaltion is essential for eukaryotic l'ell growth and changes
in this function have profound effects on cell physiology. Previous study have shown
that in the yeast S. cerevisiae, which shares the early steps of N-glyc-osylation with
higher eukaryotes, the activity of N-glycosylation function is regulated at the level
of transcription at the very first step in the pathway catalyzed by UDPGlcNAC:
Dol-P GlcNAc-Ptransferase encoded by the ALG-7 gene. To examine the
molecular controls of N-glycosyaltion and assess their involvement in development
we have studied transcriptional expression of the ALG- 7 gene in the postnatally
developing hamster salivary submandibular gland. The hamster submandibular gland
was surgically removed at 4, 12, 24, and 72 days after birth and poly-(A) + RNAs
isolated. The ALG-7 specific transcripts were quantified with a Slot blot procedure
using a fragment of hamster ALG- 7 cDNA labeled with 32P-dCTP by nick
translation. Our result indicate that at the early stages of postnatal development ,
4 and 12 days, the steady-state level of ALG-7 mRNA me ~-fold higher than in the
adult. At 24 days the level of the ALG-7 mRNA decre ases to 4-fold over the adult
level. therefore, changes in the ALG-7 expression at the level of transcription accompany postnatal development of the submandibular gland.
