Abstract
We describe the design, implementation, and limitations of a luminescence detection system for the eVOLVER parallel benchtop bioreactor system. This addition aims to expand eVOLVER's functionality, particularly in Phage Assisted Continuous Evolution (PACE) platforms by allowing real-time automated feedback control of selection pressure in PACE. This advancement holds promise for accelerating directed evolution experiments and increasing their viability. In this project we have made hardware, software, and wetware improvements to PACE on the eVOLVER (ePACE) to characterize the current capabilities of this technology. We find that while the current state of ePACE to detect luminescence signal does not have a limit of detection (LOD) viable to be used as a feedback-control mechanism, it is theoretically possible to implement the desired control mechanisms by increasing detection and/or luminescence production and significantly lowering the LOD.
